The small orange dot in front of the parameter indicates that Huygens is using default values and you will need to check/correct the values. For best results you will need to manually input the following values:

  • the z-step size for stacks
  • the time interval for time lapse imaging
  • the lens immersion refractive index (Air, Oil, Water, or Silicon (1.406))
  • the embedding medium refractive index (from manufacturer / data sheet)
  • the backprojected pinhole size (diameter) and spacing of the spinning disc (see table below)

After adding the correct values remember to click the 'Set all verified' box and 'Accept'.
