The small orange dot in front of the parameter indicates that Huygens is using default values and you will need to check/correct the values. For best results you will need to manually input the following values:
- the z-step size for stacks
- the time interval for time lapse imaging
- the lens immersion refractive index (Air, Oil, Water, or Silicon (1.406))
- the embedding medium refractive index (from manufacturer / data sheet)
- the backprojected pinhole size (diameter) and spacing of the spinning disc (see table below)
After adding the correct values remember to click the 'Set all verified' box and 'Accept'.
Backprojected Pinhole diameter and spacing (spacing is the same for both 40µm and 25µm discs):
